چکیده

DNA/RNA Description The CENPF gene structure consists of twenty exons, ranging from 92 to 3,404 bp, and nineteen introns, ranging from approximately 1 to approximately 10 kb. Transcription 10,294 bp mRNA; 9630 bp open reading frame. Pseudogene No pseudogene. Protein Description The gene encodes a protein associated with the centromere-kinetochore complex, 3210 amino acids (aa), 367594 Da, containing internal repeats, coiled-coil (potential) and NLS (potential). Expression Breast, eye, gastro-intestinal tract, heart, liver, lymph node, ovary, placenta, skin, stomach, testis. Localisation Nucleus matrix, but not in the nucleolus, reorganization to the kinetochore/centromere (coronal surface of the outer plate) and the spindle during mitosis. Function CenpF is recruited to kinetochore early in mitosis after recruitment of Bub1 and modulates kinetochore association of certain mitotic proteins including BubR1 for kinetochore assembly.

Characterization of a novel 350-kilodalton nuclear phosphoprotein that is specifically involved in mitotic-phase progression.

A gene assigned to human chromosome 1q32-41 encodes a novel protein of 3,113 amino acids containing an internal tandem repeat of 177 amino acids. The protein, which we have named "mitosin," was identified by direct binding to purified retinoblastoma protein in vitro with a region distantly related to the retinoblastoma protein-binding site of E2F-1. Mitosin is expressed throughout S, G2, and M ...

DNA topoisomerase IIα and mitosin expression predict meningioma recurrence better than histopathological grade and MIB-1 after initial surgery

BACKGROUND The 2016 WHO histopathological grade or conventional biomarker MIB-1 is insufficient for predicting meningioma recurrence after initial treatment and alternative strategies are required. In this study, we investigated whether DNA topoisomerase IIα and/or mitosin expression can predict tumor recurrence with greater accuracy than conventional methods. METHODS The expression of MIB-1,...

Mitosin (a new proliferation marker) correlates with clinical outcome in node-negative breast cancer.

Tumor proliferation rate is an important prognostic factor in breast cancer, and S-phase fraction (SPF), as measured by flow cytometry, is the most clinically validated of several methods for measuring it. However, flow cytometry is not well suited to evaluating the formalin-fixed, paraffin-embedded tumors that are routinely available or to the increasing number of small breast cancers. These a...

Unstable microtubule capture at kinetochores depleted of the centromere-associated protein CENP-F.

Centromere protein F (CENP-F) (or mitosin) accumulates to become an abundant nuclear protein in G2, assembles at kinetochores in late G2, remains kinetochore-bound until anaphase, and is degraded at the end of mitosis. Here we show that the absence of nuclear CENP-F does not affect cell cycle progression in S and G2. In a subset of CENP-F depleted cells, kinetochore assembly fails completely, t...

Frequent Amplification of CENPF, GMNN and CDK13 Genes in Hepatocellular Carcinomas

Genomic changes frequently occur in cancer cells during tumorigenesis from normal cells. Using the Illumina Human NS-12 single-nucleotide polymorphism (SNP) chip to screen for gene copy number changes in primary hepatocellular carcinomas (HCCs), we initially detected amplification of 35 genes from four genomic regions (1q21-41, 6p21.2-24.1, 7p13 and 8q13-23). By integrated screening of these ge...
